
摘要:以黄曲霉毒素B1为研究对象,制作出一种可以实现对黄曲霉毒素B1含量进行快速检测的胶体金试纸条。实验采用竞争抑制法,基于试纸条的层析作用,以胶体金为标记物与黄曲霉毒素B1单抗偶联,将黄曲霉毒素B1抗原和羊抗鼠二抗固定到硝酸纤维膜上作为检测线和质控线,制备出可以快速检测黄曲霉毒素B1的免疫层析试纸条。通过一系列的实验和优化后得出最佳条件,试纸条的检测灵敏度为20 ng/mL;精密度和准确度;与磺胺类药物、“瘦肉精”类药物均无交叉反应;试纸条在37 ℃条件下,分别在5、10、15、20后测试,效果没有明显降低。本实验制备的黄曲霉毒素B1免疫层析试纸条具有良好的精密度和灵敏度,具有良好的特异性和稳定性。本实验制备出的试纸条可以实现对生物样本中黄曲霉毒素B1的快速检测,便于实现现场快速大量检测。

Abstract:This experiment took aflatoxin
B1 as the research object, to produce a kind of colloidal gold dipstick can
realize the rapid detection of aflatoxin B1 content. Based on the chromatography effect of the test strip, colloidal
gold was used as a marker to conjugate aflatoxin B1 mab. Aflatoxin Bi antigen
and sheep anti-mouse secondary antibody were fixed on the nitrate fiber
membrane as the detection line and quality control line, and the rapid
detection of aflatoxin Bi immunochromatography test strip was prepared. After a
series of experiments and optimization, the optimal conditions were obtained. The
detection sensitivity of the test strip was 20 ng/mL. Precision and accuracy; There
was no cross reaction with sulfa drugs and clenbuteroldrugs. When the test
strips were tested at 37C after510.15 and 20 daysthe effect did not decrease
significantly. The aflatoxin B1immunochromatography strip prepared by in vivo
experiment has good precisionsensitivity, specificity and stability. The test
strip prepared in this experiment can realize the rapid detection of aflatoxin
B1 in biological samples, so as to realize the rapid and large number of
