2,5-二溴甲基对苯二甲腈及其衍生物的合成方法研究
  

目的:对制备双苯乙烯类衍生物为荧光团的双光子荧光探针的中间产物2,5-二溴甲基对苯二甲腈及其衍生物的合成条件进行研究。方法:以对二甲苯为原料,通过芳环上的溴代、亲核取代和自由基反应等过程合成2,5-二溴甲基对苯二甲腈及其衍生物,然后通过柱层析方法进行纯化并用1HNMR确认其结构。结果:对合成工艺进行适当调整得到较好的效果。在2,5-二氰基对二甲苯的合成过程中,CuCN与2,5-二溴对二甲苯的摩尔配比换成2.1:1、反应时间调整为48小时时其产率提高。在2,5-二溴甲基对苯二甲腈的合成过程中未采用火焰枪辅助加热方法,而采用较温和的紫外灯照射方法。结论:通过反应物配比、加热温度和加热方法等的适当调节,摸索到简单易行的2,5-二溴甲基对苯二甲腈及其衍生物的合成方法。

Objective: This study interested in search of synthesis condition for 2,5-bisbromomethyl-1,4-dicyanobenzene and its derivatives, which are the intermediators of preparing a series of two-photon fluorescent probe with containing distyryl benzene derivatives as the two-photon fluorophores. Methods: p-Xylene as the starting material to synthesize 2,5-bisbromomethyl-1,4-dicyanobenzene and its derivatives by a series of reaction such as bromination on benzene ring, nucleophilic substitution and radical reactions, respectively. Then purified by silica gel column and the structures were confirmed by 1H NMR. Results: Through properly adjust on synthetic process, the research achieved good results. When the reactants molar ratio of CuCN and 2,5-dicyano-p-xylene was changed with 2.1:1, and the mixtures were reacted for 48 h, the yield of 2,5-dicyano-p-xylene was increased. When the process on synthesis of 2,5-bisbromomethyl-1,4-dicyanobenzene, the researchers used a wild condition such as ultraviolet irradiation, however another paper treated with flame gun auxiliary heating method. Conclusion: The research succeed in finding simple and easy way to synthesize 2,5-bisbromomethyl-1,4-dicyanobenzene and its derivatives by adjusting the reactants molar ratio, heating temperature, and heating time, etc.
