环氧合酶3在抗炎方面的研究
环氧合酶-3(cyclooxygenase-3, COX-3),是一种来源于环氧合酶-1(cyclooxygenase-1, COX-1)的新型COX结构亚型。研究发现,COX-3主要在中枢神经系统中表达,并且在前列腺素(prostaglandin,
PG)的生物合成过程中具有重要的作用。COX-3是研究类似对乙酰氨基酚(acetaminophen)这类解热镇痛药的作用机制,以及开发新型解热镇痛药的一个重要靶点。本文对新型COX结构亚型—COX-3的分子结构、生物差异性、以及对乙酰氨基酚对COX-3的影响进行综述。
Cyclooxygenase-3(COX-3) is a novel type of cyclooxygenase, which is the key enzyme
catalyzing arachidonic acid (AA) into prostaglandin (PG). COX-3 is as a splice
variant of COX-1 with the difference that COX-3 retains an intron that is not
retained in COX-1. It discovered in many species such as canine, mouse, human
and so on. The mechanism of action of paracetamol remains unresolved. The
greater sensitivity of cells containing COX-3 to paracetamol is frequently
cited as indicating that the target of action of paracetamol is COX-3. So, the
appearance of COX-3 will be the hot spot of researching in the study of the
mechanism of acetaminophen analogues and new antipyretic analgesics. In this
paper, the molecular structure, research process, pathophysiological role and the relations with acetaminophen will be introduced.
